The lentivirus human immunodeficiency virus type 1 (HIV-1) infects peripheral blood mononuclear cells (PBMC) and productively infects and leads to disease in those cells. A recently cloned accessory viral protein, TAT, is expressed early in the viral life cycle, presumably in T cells. TAT has been implicated in the transactivation of the HIV-1 LTR and in the virulence of HIV-1 in humans. However, TAT has not been directly detected in HIV-1-infected PBMC, and its expression in HIV-1-infected cells is unknown. In this study, we found that the p10 subunit of TAT is produced by HIV-1-infected PBMC. Immunoprecipitation of TAT expressed in COS cells and HIV-1-infected PBMC with a monoclonal anti-TAT antibody and immunoblotting of the p10 protein released from these cells with a monoclonal anti-p10 antibody revealed a major band at approximately 42 kDa, consistent with the molecular weight of the TAT p10 subunit. Immunoprecipitation of TAT expressed in COS cells or HIV-1-infected PBMC with anti-p10 antibodies identified a second band at approximately 28 kDa, which was absent in cells expressing wild-type TAT, indicating that the 28-kDa protein was derived from the p10 subunit.
